What is the
P4 FPA?
The P4 FPA is a quantitative diagnostic test that uses Fluorescence Polarization (FP) technology to detect progesterone levels in milk samples of cattle. It is a simple, fast, and accurate test.
Unlike other technologies, the P4 FPA is a homogenous test conducted in the presence of alcohol. This makes the test easy to perform, consistent, and accurate.

Performance
In the graph below, the X-axis represents days post-artificial insemination. Test results on the Y-axis are provided in ΔmP units. The cutoff is 35 ΔmP. Cows with results lower than 35 ΔmP have a low progesterone level and are considered open. Note that it is easy to identify opened cows as early as day 18. You can also see that some of the pregnant cows started to show signs of heat. The P4 FPA shows 100% specificity. No pregnant cows were identified as open.
The data in this report unambiguously demonstrates the strong analytical and diagnostic performance of the milk P4 FPA developed by Ellie LLC. A specificity of 100% is critical in preventing potentially dangerous repeated insemination of already-pregnant cows. Pre-estrous cows and cows with silent estrus were also consistently identified. In tightly controlled situations (e.g., the inclusion of a synchronization protocol), the assay demonstrated high sensitivity and specificity. We expect the assay’s performance to facilitate a significant financial gain via the early detection of opened cows.

A quantitative assay that detects progesterone (P4) in milk samples of cattle. The P4 FPA is run completely in solution without binding to plastic or paper, without washing steps, and in the presence of alcohol, which helps dissolve P4 out of milk.
