Information
The TB FPA is a qualitative test that uses FLUORESCENCE POLARIZATION technology to determine the presence of antibodies against Mycobacterium bovis in bovine serum. The diagnostic test uses a fluorescein-labeled peptide with the same amino acid sequence as an epitope region of the Mycobacterium bovis bacterium MPB70 protein. TB FPA testing is performed two weeks after tuberculin testing for best results.
The TB FPA is a homogeneous, liquid-phase assay that requires no washing steps. Using portable fluorescence polarization instruments, the test can be performed in the lab or the field.
Detection of anergic animals
Antibody tests for TB are designed to detect anergic animals that do not react in gamma-interferon release assays or skin probes.
In a recent study, we tested 100 animals from a farm that was confirmed to have TB and had not started an eradication program. Of the 100 animals tested, three displayed positive results with the TB FPA but not with gamma-interferon or skin tests.
All three animals were slaughtered and showed gross lesions and advanced disease. Overwhelming TB lesions had shut down cellular immunity; therefore, the only viable method of detection was an antibody test.

The diagnostic test uses a fluorescein-labeled peptide with the same amino acid sequence as an epitope region of the Mycobacterium bovis bacterium MPB70 protein. The sequence is conserved throughout the Mycobacterium tuberculosis complex. The test is designed to detect anergic animals.
